ABSTRACT

<p><b>BACKGROUND</b>Statin therapy has affected glucose homoeostasis of type 2 diabetes patients, which could be related with bile acids metabolism. Whether bile acid metabolism and the expression of farnesoid X receptor (FXR), liver X receptor-α (LXR-α) and sterol regulatory element-binding protein (Srebp)-1c is regulated by hyperglycemia, or whether simvastatin therapy led to higher glucose is related with down-regulated expression of FXR in diabetic rats remained unclear.</p><p><b>METHODS</b>Forty male Wistar rats were randomly divided into four groups: normal control rats, insulin resistance rats, diabetic model rats, and the late simvastatin induced diabetic rats. Normal control rats were fed with standard diet, others were fed with high-fat diet. Diabetic model rats were induced by a single intraperitoneal injection of streptozotocin (STZ). The late simvastatin induced diabetic rats started simvastatin administration after STZ induced diabetic model rats. Characteristics of fasting blood glucose (FPG), lipid files and total bile acids (TBAs) were measured and the oral glucose tolerance test (OGTT) was performed after overnight fasting at the eighth weekend. RNA and protein levels of FXR, LXR-α and Srebp-1c were tested by Western blotting and reverse transcription polymerase chain reaction (RT-PCR).</p><p><b>RESULTS</b>The insulin resistance rats showed higher glucose, lipid files and lower expression of FXR compared with normal control rats (P > 0.05). The diabetic model rats showed significantly higher glucose, lipid files, TBA and lower expression of FXR compared with insulin resistance rats (P < 0.05). The late simvastatin induced diabetic rats displayed higher glucose and TBA and lower expression of FXR compared with diabetic model rats (P < 0.05).</p><p><b>CONCLUSIONS</b>Changes in bile acid homeostasis, including the alterations of bile acid levels and bile acid receptors, are either a cause or a consequence of the metabolic disturbances observed during diabetic models. Statin therapy induced hyperglycemia may be related with FXR, SHP, LXR-α and Srebp-1 pathways.</p>

ABSTRACT

This study was aimed to analyze the bioinformatics of proteomics of rat bone marrow mesenchymal stem cells (MSCs) intervened by active principle region of Yang-Xin Tong-Mai Formula (apr-YTF). The latest versions of bioinformatics tools including DAVID (http://david.abcc.ncifcrf.gov/) and GO (http://www.geneontology.org/) were combined to assign a precise function to rat bone marrow MSCs intervened by apr-YTF. KEGG and VISANT were assigned with a precise function to rat bone marrow MSCs intervened by apr-YTF. The results showed that a total of 102 biological processes were mainly involved, with 35 cellular components and 6 molecular functions. These proteins interacted in 3 signal transduction pathways. It was concluded that the following proteins and signal transduction pathways played an important role in the process of apr-YTF inducing BMSCs differentiation into cardiomyocytes. Presenilin-1 and Presenilin-2 were in the Notch signaling pathway. And syntaxin-4 protein was in soluble N-ethylmaleimide sensitive fusion protein attachment protein (SNARE). The apr-YTF played a role on MSCs from multiple sites, with multiple links through different biological processes. The bioinformatics of proteomics can predict action mechanism of traditional Chinese medicine (TCM) from the holism concept. The validation in combination with molecularbiology was a good way for TCM modernization.

ABSTRACT

Objective To investigate the risk fairs and significance of late recurrence in non-small cell lung cancer (NSCLC)patients who had undergone complete resection and remained recurrence-free for5 years. Methods 496 individuals diagnosed and surgically treated for Stage Ⅰ and Ⅱ A NSCLC were included. We examined patients at 3-month intervals for the first 2 years after surgery and typically at 6-month intervals thereafter on an outpatient basis. The follow-up evaluation included physical examination, inaging examination and tumor narkers.Pathological examination had also been performed when needed.Of these,336 patients remained recurrence-free for 5 years were followed up continuously for 3 years.Clinicopathologic factors were collected including age, sex, smoking history, preoperative serum carcinoembryonic antigen (CEA) level, surgical apprach, maximum tumor dimension on resected specimen, histologic type, histologic differentiation, intratumoral lymphatic permeation, intratumoral vascular invasion, pleural invasion and pathologic stage. The primary end event was lung cancer recurrence. The data collection ended in January 2011.Kaplan-Meier method was used for survival rate.Survival difference wasevaluated bylog-rank test.Multivariate Cox regression was used to test the relationship of recurrence-free probability to various clinicopathologic factors. Results At 5 years after resection,109 patients had died of disease and 33 had died of other causes.18 patients were alive with disease. The remaining 3 - 36 patients were alive and recurrence-free for the first 5 years. The median follow-up period of these 5-year recurrence free survivors was 27 months (range,3-36 months).34 (10.1%)patients developed a late recurrence at 3 years among the 336 patients.Recurrence was locoregional in 13 (38.2%)patients:6 patients in mediastinal lymph nodes,3 patients in ipsilateral lung,2 patients with pleural dissemination and 2 patients in other locations.Recurrence was distant in 21 (61.8%) patients:8 patients with multiple-organ metastases,13 patients with single-organ metastasis, and the latter consisted of 5 patients in contralateral lung,3 patients in liver,2 patients in central nervous system and 3 patients in other locations.Smoking history, preoperative serum CEA level, histologic differentiation, intratumoral lymphatic invasion, intratumoral vascular invasion, pleural invasion and pathologic stage were risk factors for late recurrence in univariate analysis.Multivariate Cox analysis demonstrated that intratumoral vascular invasion and intratumoral lymphatic invasion were independent risk factors for late recurrence. Conclusion NSCLC patients have a significant risk of late recurrence after complete resection 5 years, especially for the patients with intratumoral vascular invasion and intralumoral lymphatic invasion. It is needed and significant for the patients to follow up continuously after 5 years of complete resection.

ABSTRACT

BACKGROUND:During culture of bone marrow mesenchymal stem cells(BMSCs),a certain serum is commonly added in the basic medium, such as calf serum and fetal bovine serum, but there are potential biological safety risks.OBJECTIVE:To study the effects of different serum microenvironments on in vitro culture of rat BMSCs.METHODS:BMSCs were harvested from adult rat bone marrow, and cultured in vitro by whole bone marrow adherence method. The cells were cultured under the following serum microenvironment. The primary cells of autoserum group were cultured with autoserum, changing the medium with fetal bovine serum after passage. The primary calls of homogeneity foreign serum group were cultured with homogeneity foreign serum, changing the medium with fetal bovine serum after passage. The primary cells of fetal bovine serum group were cultured with fetal bovine serum, and cultured with fetal bovine serum after passage. The primary cells of Dulbecco's modified Eagle's medium(DMEM)group were cultured with serum-free DMEM,changing the medium with fetal bovine serum after passage. The morphologic changes in BMSCs were detected under an inverted phase contrast microscope.Attachment rate and growth curve were measured.Surface marker CD11b,CD45 and CD90 expression was analyzed by flow cytometry.RESULTS AND CONCLUSION:In autoserum and homogeneity foreign serum groups, the homogenicity and degrees of fusion of call morphology were improved in comparison with other two groups and the day of first passage was less than other groups. The attachment rate was greater in the autoserum, homogeneity foreign serum and fetal bovine serum groups than the DMEM group at 24,48,72 hours(P<0.01).Doubling rate was fastest in the growth curve of autoserum group, followed by homogeneity foreign serum group and fetal bovine serum group. However,no doubling phenomenon was found in the DMEM group.Flow cytometry results demonstrated that the rates of CD11b-positive and CD45-positive cells at passage 3 were above 98% under medium containing serum, and CD90-positive rate was less than 2%.We could obtain BMSCs of higher purity.However,CD11b-positive rate was 95.83%,CD90-positive rate was 2.07%,but CD45 positive rate was only 64.79% under serum-free microenvironment.BMSC purity was significantly lower under serum-free microenvironment than under serum microenvironment. Results indicated that the microenvironment of rat autoserum can improve the attachment rate, growth and purification of BMSCs.

ABSTRACT

Objective To investigate the therapeutic mechanism of the recipe of effective components from Yangxin Tongmai Formula (YTF) for myocardial ischemia by observing its effect on angiogenesis. Methods The breeding egges were divided into 6 groups: YT Ⅱ group treated with serum containing the recipe of effective components from YTF, YT Ⅰ group treated with serum containing YTF, SBW group treated with serum containing Shexiang Baoxin Pill, bFGF group treated with serum containing recombinant bovine basic fibroblast growth factor, KX group treated with blank control serum, and blank control group, 25 eggs in each group. The survival rate of chicken embryo, growth shape and vessel count of CAM model in each group were observed after 0, 24, 48, 72 and 96 hours. Results After treatment for 96 hours, the survival rate in b-FGF group and YT II group was higher than that in the other groups. The vasculature growth was rapidly in the medication group after culturing for 72 hours, and more apparent after 96 hours, especially in the b-FGF group and YT-Ⅱ group. The vessel count in b-FGF group, YT-Ⅱ group, YT-Ⅰ group and SBW group was in-creased as compared with that in KX group and blank control group (P < 0. 05), and ranged in the order as follows: b-FGF group > YT-Ⅱ group > YT-Ⅰ group > SBW group > KX group > blank control group. Conclusion YT-Ⅱ has the sim-ilar effect on improving angiogenesis as YT-Ⅰ, SBW and b-FGF, and its effect is better than that of YT-Ⅰ and SBW.

ABSTRACT

To study the relationship between the different pulse conditions and photoplethymograms, the photoplethysmogram of 432 subjects, including normal pulse condition and 10 commonly - seen abnormal pulse, was examined at Cun - kou area with a BC - 4 quantitative photoplethysmograph. Meanwhile hemodynamic parameters were analysed. As compared with the normal pulse, photoplethysmograms of different pathological pulse conditions at Cun - kou area showed their specifical parameters and the changes of hemodynamic parameters reflected features of cardiovascular pathophysiology formed by different pulse conditions. It suggests that photoplethysmograms at the Cun -kou area can supply objective basis for clinical diagnosis of different pulse conditions.
